GLP-1 medications such as semaglutide and tirzepatide carry a boxed warning about the risk of thyroid C-cell tumors. Do not use them if you or a family member has had medullary thyroid carcinoma (MTC), or if you have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). Do not use them if you are pregnant, planning a pregnancy or breastfeeding.
